Forward compatibility is the technical bridge between the current eCTD v3 and the global transition towards eCTD v4. It ensures a continuous application lifecycle, allowing the industry to move forward without duplicating content.

How the Bridge is Built: The Mechanics of Structure and Content

Forward compatibility relies on precise mapping. Think of it as a GPS coordinate system for your structure within an application. If the coordinates are off by even a single character, the connection fails, and the reviewer is left with a fragmented view of the medicinal product's history.

Historical Verification

The referenced document must already exist in the authority’s system. If a document was never successfully submitted to the regulator, it cannot serve as a valid reference.

In eCTD v4, the concept of "Context of Use" provides flexibility in the structure in ways that the current eCTD v3 is unable to. These definitions define where content sits and what it represents. To maintain forward compatibility, these must match the current v3 settings exactly; every letter and space matters.

The Lifecycle Chain

Any mismatch in keywords can break the lifecycle chain. If the system does not recognize the relationship as an exact match, a disconnect is introduced, making it a challenge for reviewers.

Structural Management

A perfect example is the European Medicines Agency (EMA), who introduced an application-level Universal Unique Identifier (UUID). This enables both the industry and EMA to offer forward compatibility seamlessly without the need for rework.

Imagine a scenario involving the Application UUID. In one instance, a submission was made using a specific UUID. Later, a duplicate was created using that same ID. Because the ID was no longer unique, the system could no longer reliably track the lifecycle.

Even a small error in how an ID is generated or reused tells the system this is a "new" or "conflicting" record. This breaks the link to the original data, making the story of the application confusing for the reviewer and potentially leading to technical queries or delays in the assessment process.
